A range of drugs that interfere with DNA replication by inhibiting the action of the topoisomerase enzymes that allow the uncoiling of the double helix-a necessary preliminary to replication. They are used as anticancer drugs and, by local application, to treat warts. Examples are topotecan (Hycamtin) and irinotecan (Campto).
