CARD16

A gene on chromosome 11q22.3 that encodes a protein which downregulates procaspase-1/CASP1 activation by inhibiting the proteolytic maturation of pro-interleukin-1 beta (IL1B) and its release in response to lipopolysaccharides during inflammation. CARD 16 induces NF-kappa-B activation during the pro-inflammatory cytokine response; it inhibits CASP1-mediated neuronal cell death, TNF-alpha, hypoxia-, UV-, and staurosporine-mediated cell death (but not ER stress-mediated cell death). It blocks CASP1 and CASP4 activation, possibly by preventing the interaction between CASP1 and RIPK2.
Molecular pathology
CARD16 is downregulated in patients with clinical Huntington’s disease.
